Abstract

Nasopharyngeal carcinoma (NPC) is a malignant tumor of epithelial cells of the nasopharynx and is a multifactorial disease that is geographically endemic in the world. The undifferentiated carcinoma is the most common type of NPC and strongly associated with Epstein-Barr virus (EBV) infection. EBV has two phases in the cycle of infection, i.e. lytic and latent phase. BARF1 latent gene is required for maintaining latent phase and the switch from latent to lytic infection. The pathogenesis of NPC is more directly reflected by carcinoma-specific viral transcriptional activity at the site of primary tumor. The aims of this study were to determine the relative expression of mRNA BARF1 EBV in NPC WHO-3 patients as a potential biomarker of NPC pathogenesis, particularly in tumor progressivity in NPC. This reseach was designed descriptively with a cross sectional study. The samples were the tissue biopsies formalin-fixed paraffin embedded (FFPE) of 22 NPC patients at Pathology Anatomy Departement, RSUD. Prof. dr. Margono Soekarjo, Purwokerto. The relative expression of mRNA BARF1 EBV was determined by RT-qPCR technique and calculated by (2-ΔCt). The relative expression of mRNA BARF1 EBV in NPC WHO3 patients was found ranging approximately from 0.708292 to 0.840177, i.e. from early stage (n = 2; 0.708841-0.712423; med = 0.710632) to late stage (n = 6; 0.708292-0.840177; med = 0,7164655). This, however, showed no significant differences statistically (n = 8; Asymp. Sig. (0,505) > 0,05). Therefore, the relative expression of mRNA BARF1 EBV has low potential as biomarker of NPC prognosis, particularly in tumor progressivity on advanced staging.
