Abstract
Barnacles genus Fistulobalanus have characteristics of complex setae on cirri III and a subsidiary tube. Sampling in this study was conducted at Tegal Beach. Taxonomic identity based on the morphological and molecular character of the genus Fistulobalanus derived from Pantai Alam Indah is not yet known, and it is necessary to see the relationship of the Fistulobalanus A1 from Pantai Alam Indah with subfamily Amphibalaninae. The purpose of this research is to know the morphology and molecular character of Fistulobalanus sp. morphotype and knowing its relationship with other species of the genus Fistulobalanus as well as Amphibalanus sister-genus. The research method used is survey method with sampling technique by purposive sampling. Morphological characters were observed by making preserved preparatory refers to Prabowo (2009). Morphological identification refers to Chant et al. (2009) and Puspasari (2001). Molecular characters were performed using the mtCO1 gene marker according to Folmer (1985) and the DNA samples were taken from the barnacle muscle tissue. The DNA samples obtained were then amplified by PCR techniques using LCO1490 and HCO2198 primers according to Folmer (1985), then sequenced by service provider laboratories to obtain DNA sequences. Morphological characters were analyzed using the Maximum Parsimony algorithm using the PAUP application, the sequenced data was verified using BLAST (Basic Local Alignment Search Tool) on the Gene Bank page, and phylogenetic analysis using MEGA 5.0 program. The results of data analysis showed that Fistulobalanus A1 morphotype coming from Tegal Beach has a general similarity of morphological characters with Amphibalanus rhizophorae with subsidairy tube and complex setae on cirri III which is the special character of the genus Fistulobalanus. Based on the molecular identification, Fistulobalanus A1 morphotype is a sister-taxa with species Amphibalanus rhizophorae with a bootstrape value of 490x with 1000 replications.
