Abstract

Actinomycetes are Gram positive bacteria that able to produce bioactive compounds e.g antifungal compound. Exploration of this bacteria has been done in terrestrial environment. Therefore, it is necessary to explore new bioactive compounds-producing actinomycetes from various environments, such as mangrove ecosystem. This research aims to know the inhibitory percentage of antifungal extract of actinomycetes against plant pathogenic fungi (Fusarium sp., Rhizoctonia solani.,Sclerotium rolfsii). This research used survey and experimental methods. Survey method was used to get the best actinomycetes isolates to against plant pathogenic fungi and to know the Rf value of antifungal compounds. Experimental method by Factorial Completely Randomized Design with two factors, i.e growth medium and incubation period. Each factor has three levels. Each treatment was repeated 3 times. The independent variables were growth medium SCN Broth (E1), ISP2 Broth (E3), ISP3 Broth (E3) and incubation period (7 days (T7), 14 days (T14), 21 days (T21), while the dependent variable was the inhibitory percentage of antifungal extract against Fusarium sp., R. solani, and S. rolfsii. The main parameter was the inhibitory percentage of antifungal extract against plant pathogenic fungi. The supporting parameters were dry biomass weight, and medium pH. The main parameters of survey method while, zone wide of actinomycetes isolate against plant pathogenic fungi, and Rf value of Thin Layer Chromatography (TLC). The data was analyzed by Analysis of Variance (ANOVA) at 95% of confidence level, then followed by Duncan's Multiple Range Test (DMRT). The result showed that 1 of 3 potential actinomycetes (SA E46 -3, SA 25, SA E40 FS) has the best capability in inhibiting the growth of pathogenic fungi, i.e actinomycetes isolate SA E40 FS. The best incubation period in producing antifungal compound was 21 days. The widest percentage inhibitory showed by treatment E1T21, with 43,38% against Fusarium sp., 41,03% against S. rolfsii, and 49,53% against R. solani. The characteristic of antifungal extract has 3 fractions of bioactive compound with Rf value ranged to 0,70-0,85.
