Abstract
Mystus gulio belongs to the Bagridae family and has the ability to live in various environmental conditions, including in water that is different salinity called eurihalin fish. Differences in conditions are factors that influence fish to be able to adapt to their environment. Environmental conditions (including chemicals, pathogens, nutrition, temperature, DO, salinity and pH) improved health and stress levels that spur fish immune responses. Changes in the number of leukocytes is one component to determine the health of fish. Differential leukocytes are one component of an attack that enters the body of the fish. The effect of differential leukocytes in this study includes neutrophils, monocytes, eosinophils and lymphocytes. This study aims to determine the status of fish kating leukocytes (M. gulio) originating from the Brugsanta Banyumas River and Cilacap Bodo River. Kating fish samples using purposive random sampling. Fish with an average weight size of 9.16 ± 1.63 g and a day averaging 10.06 ± 1.19 cm. Kating fish was found from two places with different salinity, namely in Brugsanta River Sirau Village, Kemranjen District, Banyumas Regency with salinity 0 ppt and Bodo River Jetis Village, Nusawungu District, Cilacap Regency with salinity 24-35 ppt. Data obtained be analyzed using a t test to compare differential leukocytes data of fish from two sampling locations. The variables used in this study are independent variables and variables. The independent variable in this study is location. The variable in this study is differential leukocytes. The parameters used are differential leukocytes percentage (including lymphocytes, monocytes, neutrophils, and eosinophils). The observation of kating fish leukocytes originating from Brugsanta River, Banyumas didn’t different significantly (P>0.05) with differential leukocytes fish at Bodo River, Cilacap. The percentage of kating lymphocytes from Brugsanta, Banyumas and Kali Bodo, Cilacap were 67.96 ± 2.81% and 69.05 ± 3.42%, the percentage of monocytes 25.91 ± 2.79% and 25.18 ± 3.38%, neutrophil percentage 5.75 ± 1.13% and 5.32 ± 1.11%, and the percentage of eosinophils was 0.38 ± 0.49% and 0.45 ± 0.49%.
