Abstract
Shiitake mushrooms (Lentinula edodes) and oyster mushrooms (Pleurotus ostreatus) is edible mushrooms and medicinal mushrooms because they contain high nutrients and bioactive compounds, such as flavonoids, phenols and terpenoids that can function as antioxidants. Antioxidants are molecules that are able to ward off free radicals in the body, although naturally the body can ward off free radicals through enzymatic (endogenous) antioxidants, such as superoxide dismutase (SOD) and catalase. But if too many free radicals enter the body, it can cause oxidative stress so that endogenous antioxidants cannot work properly, so exogenous antioxidants are needed such as those found in ethanol extracts of L. edodes and P. ostreatus. One of the materials that can become free radicals is carbon tetrachloride (CCl4). This study aims to determine the effect of ethanol extract of L.edodes and P. ostreatus on levels of SOD, catalase in rats and determine the most effective dose of ethanol extract on increasing levels of SOD and blood catalase in rats induced by CCl4 and determine the antioxidant activity and IC50 value of L. edodes and P. ostreatus. This research carried out experimentally with a Completely Randomized Design (CRD) with two treatments, namely the type of mushroom (L. edodes and P. ostreatus) and variation of ethanol extract concentration (250; 500; 750 ppm). In this study consisted of 9 treatments with 4 replications to obtain 36 experimental units consisting of K+ as a positive control, K-as a negative control, KP as a comparison group, P1 treatment induced CCl4 and extract ethanol of L. edodes at a dose of 250 mg/kg BW, P2 treatment induced CCl4 and extract ethanol of L. edodes at a dose of 500 mg/kg BW, P3 treatment induced CCl4 and extrac ethanol t of L. edodes at a dose of 750 mg/kg BW, P4 treatment induced CCl4 and extract ethanol of P. ostreotus at a dose of 250 mg/kg BW, P5 treatment induced CCl4 and extract ethanol of P. ostreotus at a dose of 500 mg/kg BW, P6 treatment induced CCl4 and extract ethanol of P. ostreotus at a dose of 750 mg/kg BW. The parameters of this study were IC50 value, sod and catalase levels. The research data obtained were then analyzed using Analysis of Variance (ANOVA) with an error rate of 5%. The results showed that the extraction of shiitake mushroom extract and oyster mushroom has an effect on increase in the levels of superoxide dismutase and catalase in rats induced carbon tetrachloride. The concentration of ethanol extract of 500mg/kg BW of rats was the most effective concentration for increasing superoxide dismutase and catalase and the ethanolic extract of L. edodes and P. ostreotus had antioxidant activity. The IC50 value of L. edodes ethanol extract was 36.02 mg/ml which was classified as very strong and the IC50 value of P.ostreotus ethanol extract was 67.02 mg/ml which was classified as strong.
