Abstract

Latar Belakang: Kanker telah menjadi penyebab kematian terbanyak kedua di dunia dan penanganannya menghadapi beberapa tantangan seperti resistensi, tidak selektif, dan efek samping kemoterapi. Di sisi lain, eksploitasi sumber daya alam yang berlebihan dalam penemuan obat baru untuk terapi kanker dikhawatirkan dapat membahayakan alam atau menyebabkan kepunahan tanaman. Oleh karena itu, para peneliti mulai mencari alternatif sumber yang kaya akan senyawa bioaktif diantaranya jamur endofit. Metodologi: Jamur endofit diisolasi dari tanah tempat tumbuh lempuyang gajah hingga diperoleh isolat jamur tunggal dan murni. Isolat jamur kemudian diidentifikasi secara molekuler menggunakan teknik Polymerase Chain Reaction (PCR). Fermentasi jamur dilakukan pada media padat kacang hijau dan beras sebagai media standar, serta kacang hijau tanpa ditumbuhi jamur sebagai kontrol media. Ekstraksi dilakukan dengan etil asetat (EtOAc) menggunakan metode maserasi dengan penggojokan (shaking). Analisis fitokimia dilakukan dengan metode KLT dan KCKT. Uji toksisitas dilakukan terhadap larva udang (Artemia salina) dengan metode Brine Shrimp Lethality Test (BSLT), dilanjutkan analisis probit untuk memperoleh nilai LC50. Hasil Penelitian: Jamur endofit yang berhasil diisolasi dari tanah tempat tumbuh lempuyang gajah teridentifikasi secara molekuler sebagai Mucor circinelloides. Ekstrak EtOAc jamur M. circinellloides pada media kacang hijau menunjukkan keberadaan senyawa flavonoid, sedangkan pada media beras menunjukkan adanya senyawa terpenoid atau steroid berdasarkan profil KLT. Analisis secara KCKT mengindikasikan adanya kemiripan pola spektrum UV senyawa pada ekstrak jamur dalam media kacang hijau dengan ekstrak kontrol media serta ekstrak jamur dalam media standar pada tR yang berbeda. Pada uji BSLT, ekstrak EtOAc jamur M. circinellloides yang dikultur pada media kacang hijau dinyatakan toksik dengan nilai LC50 sebesar 758,58 ppm. Kesimpulan: Jamur endofit M. circinelloides yang telah diisolasi dari tanah dan difermentasikan dalam media kacang hijau mengandung senyawa flavonoid dan berpotensi toksik terhadap larva A. salina. Background: Cancer has become the second leading cause of death in the world and its handling faces several challenges such as resistance, non-selectiveness, and side effects of chemotherapy. On the other hand, the excessive exploitation of natural resources in drug discovery for cancer is treatening the biodiversity and even causing plants extinction. Accordingly, some scientists try to explore untapped sources for bioactive compounds including endophytic fungi. Methods: A single and pure endophytic fungus was isolated from soil sample around the habitat of Zingiber zerumbet. Subsequently, this fungus was molecularly identified by using Polymerase Chain Reaction (PCR). The fungus fermented on solid media of mung beans and rice as a standard media, in addition to fermentation of mung beans without fungus as a control media. Extraction was carried out with ethyl acetate (EtOAc) using maceration method with shaking. Phytochemical analysis was carried out by using TLC and HPLC methods. Toxicity test was performed on Artemia salina larvae using the Brine Shrimp Lethality Test (BSLT) method, followed by probit analysis to determine an LC50 value. Results: An endophytic fungus isolated from the soil sample was molecularly identified as Mucor circinelloides. Based on the TLC profile, EtOAc extract of M. circinellloides on mung bean media indicated the presence of flavonoids, while its fermentation on solid rice media showed the presence of terpenoids or steroids. HPLC analysis indicated that there were similarities in the UV spectrum of compounds in fungi extract on mung beans media with control media extract and fungi extract on standard media at different tR. In the toxicity screening, EtOAc extract of M. circinellloides on mung bean media revealed toxicity against A. salina larvae with LC50 value of 758.58 ppm. Conclusion: The soil-derived endophytic fungus M. circinelloides which has been fermented on mung beans media which contains flavonoids and showed potential toxicity against A. salina.
