Abstract
Acute respiratory infections (ARIs) are all infections in the upper and lower respiratory tract. Paramyxovirus, which is also referred to Paramyxoviridae family viruses, contributes to ARI cases and reported to have a high human-to-human transmission rate, especially in children. The detection of Paramyxovirus by molecular technique, one of which is Nested Reverse Transcription PCR (RT-PCR) can be used to diagnose ARIs and improve the efficiency of ARI examinations. Puskesmas Batur I and II from January to August 2023 reported 24.9% ARI cases in children with a total of 5,105 cases. Therefore, the Batur area was chosen as a research site to identify the viral etiology of ARI cases in children. This study is expected to detect Paramyxovirus using Nested RT-PCR technique and provide data based on the calculated prevalence rate of Paramyxovirus infection cases in children diagnosed with ARI in Batur District, Banjarnegara Regency. This study was conducted as descriptive study in the form of a cross-sectional study and survey method with purposive sampling. The research sampling was taken by nasopharyngeal (NP) swab from children with ARI located in Puskesmas Batur I and Batur II, Batur District, Banjarnegara Regency. The research method used Nested Reverse Transcription PCR (RT-PCR) method with electrophoresis for the analysis detection and Sanger DNA sequencing for verification of the amplified PCR product. The results of this study showed that Nested RT-PCR in this study detected the presence of Paramyxovirus at family-level in children with ARI nasopharyngeal swab samples. Therefore, the calculated prevalence of Paramyxovirus at family-level in children with ARI cases within the Batur District are 25% which considered to be high.
