Abstract
Padina australis Hauck is one of the seaweed species of the Phaeyophyta division which is commonly spread in marine waters, both shallow sea waters to deep waters. This seaweed has a form of talus in the form of a wide sheet of brown transparent. This seaweed is brown because in its talus contained fukosantin pigment and xantofil. In addition to fukosantin, this seaweed also has chlorophyll a and c, fukosantin and chlorophyll are present in the talus plastid. Studies on the anatomical structure of seaweed, especially P. australis so far have not been much studied. Anatomically the constituent talus consists of three types of cells from the inside out ie, epidermal cells, cortex and medulla. Talus of seaweed has reproductive organs whose shape and structure are different depending on sex and stadia. The reproductive organs of P. australis are called tetrasporangia an organ containing tetraspores. The location of tetrasporangia P. australis is found in the concentric line. Each talus of P. australis has a talus size and the number of varied concentric lines. The aim of this research is to know the anatomical character of talus shape of P. australis based on the concentric line and to know the difference of anatomical character of P. australis based on the number of concentric lines and the talus section. Seaweed sampling of P. australis by survey method with purposive random sampling technique is taking samples of randomly selected seaweed which is found in Karang Tengah beach, Cilacap regency. Preparation of microscopic by paraffin method or embedding staining with safranin 1% in 70% alcohol. Parameters observed included thickness of epidermis, size of medullary cell, thickness of talus and size of tetraporangia. Observation of anatomical structure is descriptively by comparing the anatomical character of P. australis Hauck on the concentric line. The anatomical character data was analyzed by t-Test with 95% confidence level. Result of the research showed that the anatomical character of talus P. australis with concentric line 5 had a thickness of upper and lower epidermis respectively is 20.05 μm and 12.55 μm, whereas in concentric line 7 had a thickness of 28.33 μm and 18 μm. The thickness of the talus with concentric line 5 and 7 respectively is 99.44 μm, 114.77 μm. Medullary diameter with concentric line 5 and 7 respectively is 37,88 μm, 45,5 μm. Average diameter of tetrasporangia with concentric line 5 and 7 respectively that is 25,66 μm, 35,66 μm. Result of analyzed by t-Test showed that the talus P. australis based on concentric lines 5 and 7 has differences of sizes on the upper epidermal thickness, thickness of the lower epidermis, thickness of talus, medullary diameter and diameter of tetrasporangia.
